During week seven in Genetics, students need:
2 LB/Tobramycin plates
3 LB/Tobramycin tetracycline plates
1 uracil drop-out plate
1 YPD plate
a set of controls that include:
LB
LB/tet
LB/tob
LB/tet/tob

| One section's plates coming out of the incubator |
This does not include the yeast and bacteria controls, previous plates and a solid TON of flat-edged toothpicks, OR the .1% Sodium Pyrophosphate
So what it mean is:
11 plates x 6 groups x 13 labs, OR 858 (or THREE CASES) of plates all hand-pour by two work study students (with TERRIBLE schedules this week) or... ME.
In Micro, it is concurrently, PCR week which means.
ordering primers
reconstituting said primers
aliqoting primers, enzyme, master mix
setting up broth tubes for every group (x6)
Then setting up 6 gel boxes,
making enough TAE for all sections
setting up ethidium bromide station
and making sure gel documentation station is fine
This on top of week one of real gene annotation (remember, this is all new to me), my "super" Tuesday that goes like this:
Arrive Burrus to make sure the 8 am Genetics lab is ok: 8:45
Set out freezer components of PCR for micro and assure brand-new instructor he will be fine 9:00
Get in car, rush to ISAT for Viral Discovery 9:15-10:45
Rush BACK to Burruss for office hours 11-12
rush BACK to ISAT for Viral Discovery lab12:01
third floor for samples
first floor to centrifuge them
third floor to open lab, see note
first floor to get samples
third floor to tell game plan
first floor to get students' samples out of centrifuge
lab 12:20-1:50
back to Burruss 2:00
My section of Genetics 2:30-5:30
Needless to say, I have had quite a week.

Perfect gel after perfect gel... well for most of the groups. Not bad. I deserve a special treat.
